Class B (type K) CpG oligonucleotides - Stimulatory human/mouse TLR9 ligands
Class B (also known as type K) CpG ODNs (CpG-B ODNs) are synthetic TLR9 agonists composed of a fully phosphorothioate-modified backbone and one or more unmethylated CpG motifs. The optimal motif is GTCGTT in humans and GACGTT in mice. They strongly activate B cells, promoting proliferation, antibody production, and pro-inflammatory cytokine release (e.g., IL-6, TNF-α). Unlike Class A, they weakly stimulate IFN-α secretion from plasmacytoid dendritic cells. Class B CpG ODNs are widely used in vaccine adjuvant research, cancer immunotherapy, and B cell function studies.
InvivoGen provides seven known Class B CpG ODNs:
- ODN 1668: 5’-tccatgacgttcctgatgct-3’ (20 mer)
- ODN 1826: 5’-tccatgacgttcctgacgtt-3’ (20 mer)
- ODN 2006 (ODN 7909): 5’-tcgtcgttttgtcgttttgtcgtt-3’ (24 mer)
- ODN 2007: 5’-tcgtcgttgtcgttttgtcgtt-3’ (22 mer)
- ODN BW006 (ODN 684): 5’-tcgacgttcgtcgttcgtcgttc-3’ (23 mer)
- ODN D-SL01: 5’-tcgcgacgttcgcccgacgttcggta-3’ (26 mer)
- ODN CpG-B (VacciGrade™️):5′-tgactgtgaacgttcgagatga-3′ (22 mer)
