Human IL-1β protein - Mammalian cell-expressed, tag-free, carrier-free

Recombinant human IL-1β is a high-quality and biologically active cytokine, validated using proprietary IL-1β reporter cells. This member of the IL-1/TLR cytokine superfamily is produced in CHO cells to ensure protein glycosylation and bona fide 3D structure.

Recombinant human IL-1β can be used together with HEK-Blue™ IL-1β cells for the screening of inhibitory molecules, such as Canakinumab, a therapeutic monoclonal antibody targeting IL-1β (see figures).

Interleukin 1 beta (IL-1β) is a secreted pro-inflammatory cytokine that generates systemic and local responses to infection and injury. Dysregulated production of IL-1β contributes to inflammatory diseases such as cryopyrin-associated periodic syndrome (CAPS), familial mediterranean fever (FMF), and systemic juvenile idiopathic arthritis (SJIA).

SDS PAGE of the recombinant human (h)IL-1β protein. 3 μg of CHO-derived hIL-1β was loaded on a 12% Mini-PROTEAN® TGX Stain-Free™ Precast Gel (Bio-Rad). Detection was performed as per the manufacturer’s instructions. Its predicted size is ~17 kDa (lower band). The higher band at ~19 kDa corresponds to the glycosylated protein.

Dose-response in HEK-Blue™ IL-1β cells to recombinant IL-1β cytokines of different sources. Cells were stimulated with increasing concentrations of recombinant human IL-1β, derived from E. coli (blue) or CHO cells (red). After overnight incubation, the NF-κB-induced SEAP activity was determined using QUANTI-Blue™, a SEAP detection reagent. Data are shown as optical density (OD) at 650 nm (mean ± SEM).

Anti-hIL-1β-IgG is a potent neutralization antibody. Increasing concentrations of Anti-hIL-1β-IgG were incubated for 30 minutes with the recombinant cytokine hIL-1β (1 ng/ml) prior to the addition of HEK-Blue™ IL-1β cells. After overnight incubation, SEAP activity in the cell culture supernatant was assessed using QUANTI-Blue™ Solution. Data are shown in percentage (%) of neutralization.

Details

IL‑1 is a secreted pro‑inflammatory cytokine that plays a critical role in immune responses and inflammation [1]. It exists in two distinct isoforms, IL-1α and IL-1β, which are produced as pro-proteins by activated macrophages. IL-1β is cleaved by caspase-1 [2] while IL-1α is cleaved by calcium‑activated calpain or caspase-5 (or its murine ortholog caspase-11) [3]. In contrast to pro‑IL‑1β, pro-IL-1α is active, although cleavage dramatically enhances its bioactivity. IL-1β and IL-1α bind to the same receptor, IL-1R1, triggering the formation of the IL-1R1/IL-1R3/MyD88 complex. This induces signaling leading to the activation of the transcription factors NK-κB and AP-1 with the subsequent inflammatory response [4].
