Secreted ACP5 Reporter Gene in expression plasmid
InvivoGen has developed a secreted version of the acid phosphatase 5 (ACP5) that can be used as a reporter gene. ACP5, also known as tartrate-resistant acid phosphatase (TRAP or TRAPase) is a metalloprotein enzyme, usually located in the lysosome.
Description
InvivoGens secreted (s)ACP5 features a signaling sequence, that - when expressed in mammalian or insect cells - mediates the export of the sACP5 protein from within the cell to the surrounding culture medium. The level of ACP5 activity in the cell culture supernatant is easily quantified using QUANTI-Star™, our easy-to-use detection reagent.
Within the pUNO1 plasmid, the sACP5 gene expression is driven by the EF-1α/HTLV composite promoter that combines the elongation factor 1 alpha core promoter and the 5’untranslated region of the Human T-cell Leukemia Virus. The pUNO1-sACP5 plasmid is selectable with Blasticidin in both mammalian cells and bacteria. The gene is flanked by convenient cloning sites for easy subcloning (5’ end: Agel, NcoI. 3’ end: EcoR I site).
The pUNO1-sACP5 plasmid can be subcloned downstream of any promoter of your choice and used in vivo and in vitro to transfect mammalian cells stably or transiently.
