Genetic adjuvant for DNA vaccines featuring the murine mFLT3L and GM-CSF genes
pBOOST4-mFLT3L-mGMCSF plasmid encodes the murine Fms-like tyrosine kinase 3 ligand (FLT3L) and granulocyte-macrophage colony-stimulating factor (GM-CSF).
FLTL3L is a synergistic hematopoietic growth factor that mediates cell survival, cell proliferation, and differentiation of dendritic cells [1]. GM-CSF, also known as colony-stimulating factor 2 (CSF2), is an important hematopoietic growth factor and immune modulator [2]. Mouse co-immunization with both FLT3L and GM-CSF encoding plasmids has been reported to increase antibody titer up to 9-fold [3].
Murine FLT3L and GMCSF genes are under the control of two strong composite promoters derived from the ferritin heavy chain (hFerH) and light chain (hFerL) core promoters, respectively.
pBOOST4-mFLT3L-mGMCSF plasmid is selectable in E. coli with Blasticidin.
Note: Murine FLT3L and GMCSF genes are also available in two separate plasmids, pUNO1-mFLT3LGa and pUNO1-mGMCSF.
