Coelenterazine-utilizing luciferase detection reagents

QUANTI-Luc™ 4 is a liquid ready-to-use bioluminescent assay reagent to measure the activity of Lucia luciferase and other coelenterazine-utilizing luciferases. In order to meet your practical needs, InvivoGen offers:

— QUANTI-Luc™ 4 Lucia/Gaussia: for the detection of secreted Lucia and Gaussia luciferases 
— QUANTI-Luc™ 4 Renilla: for the detection of intracellular Renilla luciferase — NEW !

 

Lucia, Gaussia, and Renilla luciferases catalyze the oxidation of their coelenterazine substrate with the emission of light (bioluminescence). The light signal produced correlates to the amount of luciferase protein expressed. It is quantified using a luminometer and expressed as relative light units (RLUs).

Our proprietary QUANTI-Luc™ 4 Reagent has been optimized to reduce the coelenterazine substrate auto-luminescence for a better detection range. The optimal pH value for maximal luciferase activity differs between Lucia, Gaussia, and Renilla. The final pH of the reaction using our reagents confers one type of luminescence detection for Renilla and two types for Lucia/Gaussia (see figures):

Flash luminescence emits light with high intensity for a few seconds. It is ideal for end-point detection in small sample numbers, or when using microplate readers with reagent injectors.
Glow luminescence emits light with stable intensity for over 10 minutes. It is ideal for high-throughput screening (HTS) (i.e. the typical duration to read a 1536-well plate is ~7 minutes) or when using a luminometer without injectors (end-point detection).

Lucia luciferase detection over 10 minutes using QUANTI-Luc™ 4 Lucia/Gaussia detection solution. THP1-cells, which feature an IRF-inducible Lucia luciferase reporter gene, were incubated with 30 µg/ml of 2'3'-cGAMP, a STING agonist. After overnight incubation, the Lucia activity in the supernatant was assessed using QUANTI-Luc™ 4 Lucia/Gaussia without or with stabilizer to measure the flash and glow luminescence. Data are shown as (A) relative light units (RLUs) or (B) fold change over non-induced cells.

End-point detection of Lucia luciferase. A549 cells stably transfected with an IRF-inducible Lucia reporter gene were cultured with increasing concentrations of 3p-hpRNA complexed with LyoVec. After overnight incubation, the Lucia activity in the supernatant was assessed immediately after the addition of InvivoGen's QUANTI-Luc™ 4 Lucia/Gaussia (without stabilizer). Data are shown as relative light units (RLUs).

Renilla luciferase detection for 20 minutes using QUANTI-Luc™ 4 Renilla. HEK 293 cells were stably transfected with an NF-κB-inducible plasmid encoding the wildtype Renilla luciferase. These cells were incubated with 10 ng/ml of hTNF-α. After overnight incubation, the cells were lysed using QUANTI-Luc™ 4 lysis buffer and the Renilla activity was measured using the QUANTI-Luc™ 4 Renilla detection reagent. Data are shown as (A) relative light units (RLUs) or (B) fold change over non-induced cells.

QUANTI-Luc™ 4 Lucia/Gaussia

Cost-effective: One unit is sufficient for 5 x 96-well plates or 2 x 1536-well plates.

Practical: After preparation, QUANTI-Luc™ 4 Lucia/Gaussia is stable for 48 hours at 4°C and 1 month at -20°C.

Quality control: Each lot is thoroughly tested in functional assays using the recombinant Lucia protein or Lucia reporter cells. The detection of constitutive or inducible Gaussia has been verified using cellular assays.

 

QUANTI-Luc™ 4 Renilla

Quality control: The detection of constitutive or inducible Renilla has been verified using cellular assays.
