TLR7-DN (ΔTIR) : Dominant Negative TIR-less TLR7 gene

 

TLR7 is phylogenetically close to TLR8 and TLR9 and has a higher molecular weight when compared with hTLR1-6, largely as a result of a longer ectodomain [1]. TLR7 recognizes single stranded RNAs and small synthetic molecules such as imidazoquinolines and nucleoside analogs [2,3].
TLR7-DN is a TIR-less form of the TLR7 gene generated by deleting the TIR domain (480 bp). This truncated TLR7 gene is still able to recognize its ligands but is unable to induce the signaling cascade. The gene has been fused at the 3'end to the influenza hemagglutinin (HA) tag. This short sequence (YPYDVPDYA) encodes a peptide which is the epitope of a very efficient and specific monoclonal antibody. The use of HA-tagged genes provides a simple and convenient method to detect the expression of the genes by Western blot.
