TICAM1 (TRIF) is an adapter involved in the MyD88-independent signaling pathway of several TLRs. A dominant-negative form of TICAM1 was shown to completely abolish TLR3-induced signaling and also to block the NF-kB activity mediated by TLR2, TLR4, and TLR7 [1]. Further analysis has revealed that TICAM1 binds preferentially to TLR3 and TLR4 and is essential in their signaling pathways [2]. It predominantly activates IRF-3 and the production of IFN-b rather than NF-kB [3].
