Pellino homolog 1 (Drosophila).
Pellino is a Pelle-interacting protein recently identified in Drosophila. So far three members of the Pellino family have been identified in mammals. Pellino1, Pellino2 and Pellino3 share high amino acid identity. Pellino1 is required for IL-1-induced NF-kB activation and IL-8 gene expression [1]. Pellino2 interacts with IRAK-1 and IRAK-4 and activates the MAP kinase pathway leading to activation of AP-1 and Elk-1 [2]. Pellino3 participates in the activation of c-Jun and Elk-1, but not NF-kB [3]. Thus the Pellino proteins are intermediate components of the IL-1R/TLR signaling pathways that act as scaffolding proteins and may regulate signaling branch points.
