Class C CpG oligonucleotides - Stimulatory multispecies TLR9 ligands
Class C CpG ODNs are synthetic TLR9 agonists that combine structural and functional features of Class A and Class B CpG ODNs. They are characterized by an entire phosphorothioate backbone and palindromic CpG motifs capable of forming secondary structures. They induce high IFN-α production from plasmacytoid dendritic cells (like Class A CpGs) and strongly activate B cells, promoting proliferation, antibody production, and pro-inflammatory cytokine release (like Class B CpGs). Class C CpG ODNs are used as potent vaccine adjuvants, in cancer immunotherapy, and in infectious disease research. They are also widely used in in vitro TLR9 pathway studies.
InvivoGen provides three known Class C CpG ODNs:
- ODN 2395: 5’- tcgtcgttttcggcgcgcgccg-3’ (22 mer)
- ODN D-SL03: 5’- 5’-tcgcgaacgttcgccgcgttcgaacgcgg-3’ -3’ (29 mer)
- ODN M362: 5’-tcgtcgtcgttcgaacgacgttgat-3’ (25 mer).
Note: Regular letters represent phosphorothioate bonds, bold letters represent CpG dinucleotides, underlining indicates palindromic sequence.
