Receptor interacting serine/threonine kinase 3.
Receptor-interacting protein 3 (RIP3) is a member of the RIP Ser/Thr kinase family [1]. Unlike RIP1 and RIP2, RIP3 possesses neither a death domain nor a CARD motif at its unique C terminus [2]. RIP3 is a potent inducer of apoptosis, an activity readily localizable to its C terminus. RIP3 binds RIP within the TNFR-1 signaling complex and attenuates its ability to engage the NF-kB pathway [2,3]. Upon TNF stimulation, RIP1 is recruited to the TNFR1 signaling complex to activate NF-kB. RIP3 is then recruited and phosphorylates RIP1 inhibiting its ability to activate NF-kB. Thus RIP3 acts as a modulator that can limit persistent activation of the NF-kB pathway [3].
