Myeloid Differentiation Primary Response 88 (MyD88)
Myeloid differentiation factor 88 (MyD88) is a cytoplasmic adaptor protein that is involved in interleukin-1 receptor (IL-1R)- and Toll-like receptor (TLR)-induced activation of NF-kappaB. MyD88 contains a C-terminal Toll/IL-1R homology (TIR) domain and an N-terminal death domain (DD), which mediate the interaction of MyD88 with IL-1R/TLR and IRAK, respectively [1]. MyD88 is essential for signaling as MyD88 -/- mice fail to induce cytokines in response to LPS and CpG DNA [2]. Some TLRs induce antiviral responses by producing IFN-alpha in a MyD88-dependent manner. MyD88 interacts with IRF7 resulting in the activation of IFN-alpha-dependent promoters [3].
