Class A CpG oligonucleotide - Stimulatory human TLR9 ligand
InvivoGen provides ODN 2216, a Class A CpG oligonucleotide (ODN) with a preference for the human Toll-like receptor 9 (TLR9). CpG Class A ODNs, such as ODN 2216, are characterized by a phosphodiester (PO) central CpG-containing palindromic motif and a phosphorothioate (PS)-modified 3’ poly-G string.
CpG ODNs are synthetic oligonucleotides that contain unmethylated CpG dinucleotides in specific sequence contexts (CpG motifs). These CpG motifs are present at a 20-fold greater frequency in microbial DNA than in mammalian DNA. CpG ODNs are recognized by TLR9, which leads to strong immunostimulatory effects.
The sequence of the ODN 2216 is 5’-ggGGGACGA:TCGTCgggggg-3’ of 20 mer. The bases shown in capital letters are phosphodiester, and those in lowercase are phosphorothioate (nuclease resistant). The palindrome is underlined.
